PEG-MGF Dosage Protocol

PEGylated mechano growth factor · IGF-1 splice-variant analogue

The reference protocol starts at 200 mcg once daily for weeks 1–2, then increases by 100 mcg every 2 weeks to 500 mcg in weeks 7–8. With 3 mL of bacteriostatic water, a 2 mg vial is 0.667 mg/mL, so 200 mcg is 30 units (0.30 mL) on a U-100 syringe. It is a research-only protocol, not an established human dose.

02 · HOW TO MIX

Mixing a 2 mg vial with 3 mL of BAC water

Using a 1 mL (100-unit) insulin syringe, 3 full draws of bacteriostatic water gives 0.67 mg/mL.

  1. Bacteriostatic water running slowly down the inside wall of a peptide vial1

    Add the water slowly

Wipe both rubber stoppers with an alcohol swab and let them dry. Draw 3 mL of bacteriostatic water and let it run slowly down the inside of the vial wall — not straight onto the powder.

  2. Hand gently swirling an upright vial in a small circle2

    Swirl gently

Swirl or roll the vial gently until the powder has dissolved. Give it a few minutes to settle if needed.

  3. A vial being shaken with a red cross through it3

    Don't shake it

Shaking can damage the peptide and cause foaming. Gentle swirling only.

  4. Close-up of a vial with small undissolved particles and a caution symbol4

    Check it's fully clear

Only use it once it is fully clear with no particles. Label the vial with the date and 0.67 mg/mL, then store it in the fridge at 2–8 °C.

How it works

PEG-MGF is marketed as a PEGylated version of mechano growth factor, an IGF-1 gene splice variant also called IGF-1Ec. PEGylation is intended to alter how long a molecule remains in the body, but this does not establish the behaviour or benefit of a PEG-MGF product in people.

In human muscle biopsies after exercise-induced damage, MGF mRNA rose rapidly and transiently. Cell and animal experiments suggest synthetic MGF E-peptide analogues can affect muscle progenitor cells, but their receptor, processing and wider effects remain uncertain.

Illustrated mechanism explainer

What the research shows

  • In a study of 10 healthy men with exercise-induced muscle damage, MGF mRNA rose rapidly and transiently; the signalling tests in that report were done in C2C12 muscle-like cells, not with PEG-MGF dosing in people.
  • A primary-cell experiment found that the 24-amino-acid MGF E peptide increased proliferative lifespan in neonatal and young-adult human muscle-cell cultures, but not in cells from older adults. This was not a PEG-MGF clinical study.
  • In a 2019 mouse injury model, MGF injection did not protect muscle-fibre regeneration; it reduced fibrosis in the macrophage-depleted injury model. Molecular analyses used groups of 8 mice and regeneration analyses used groups of 4 mice.
  • The TGA ARTG public search for PEG-MGF returned no matching results. PEG-MGF is not an approved medicine in Australia, and no FDA-approved PEG-MGF label was identified.
  • A review concluded that endogenous short MGF E peptides had not been confirmed and that the mechanism and wider biological effects of these synthetic peptide mimetics remained incompletely understood.

Reported side effects

  • Human safety data for injected PEG-MGF at the chart amounts have not been established, so the frequency and severity of adverse effects are unknown.
  • In 2 black-market MGF preparations analysed by mass spectrometry, the products were C-terminally amidated analogues rather than the reference human MGF sequence, illustrating an identity and quality risk in unregulated products.
  • Growth-factor signalling and the broader biological effects of MGF E-peptide mimetics remain uncertain; cell and animal findings cannot establish safety in people.

Important notes

  • No PEG-MGF medicine was found in the TGA ARTG public search. It is not approved for human therapeutic use in Australia.
  • Prohibited in sport: WADA lists IGF-1 and its analogues, plus other growth factors or modulators affecting muscle or regenerative processes, under S2 at all times. Athletes should obtain advice from their anti-doping organisation.
  • There is no clinically established human dose for PEG-MGF. The chart copies the attributed educational reference protocol rather than a medicine label or human dose-escalation study.
  • Unregulated MGF products may not contain the claimed molecular form; a published analysis found amidated MGF analogues in 2 black-market preparations.
  • Never use a mixed vial that is cloudy or has particles in it.

Injection basics

  • Use a new sterile 1 mL (100-unit) U-100 insulin syringe, 27–31G, every time — never reuse or share.
  • Wipe the vial stopper and the skin with an alcohol swab and let both dry first.
  • Inject under the skin (subcutaneously) of the abdomen, thigh or upper arm, and rotate sites.
  • Put used syringes straight into an approved sharps container — the NDSS lists Australian disposal options.

06 · QUICK ANSWERS

PEG-MGF questions

How many units is 200 mcg of PEG-MGF in a 2 mg vial?

With 3 mL in a 2 mg vial, the concentration is 0.667 mg/mL. A 200 mcg amount is 30 units, or 0.30 mL, on a U-100 syringe.

How much bacteriostatic water is used for PEG-MGF 2 mg?

The attributed 2 mg protocol uses 3 mL, giving 0.667 mg/mL. At that concentration, 300 mcg is 45 units (0.45 mL), 400 mcg is 60 units (0.60 mL), and 500 mcg is 75 units (0.75 mL).

How long does a 2 mg PEG-MGF vial last?

At 200 mcg once daily, a 2 mg vial provides 10 doses. At 300 mcg it provides about 6.7 doses, at 400 mcg 5 doses, and at 500 mcg 4 doses.

Is PEG-MGF banned in sport?

WADA prohibits IGF-1 and its analogues, and also other growth factors or growth-factor modulators affecting muscle or regenerative processes, under S2 at all times.
