MGF Dosage Protocol

Mechano growth factor · IGF-1 splice-variant research peptide

The reference protocol starts at 100 mcg once daily in week 1, then rises weekly to 150, 200, 250 and 300 mcg for weeks 5–8. With 3 mL of bacteriostatic water, a 5 mg vial is 1.67 mg/mL, so 100 mcg measures 6 units (0.06 mL) on a U-100 syringe.

02 · HOW TO MIX

Mixing a 5 mg vial with 3 mL of BAC water

Using a 1 mL (100-unit) insulin syringe, 3 full draws of bacteriostatic water gives 1.67 mg/mL.

  1. Bacteriostatic water running slowly down the inside wall of a peptide vial1

    Add the water slowly

Wipe both rubber stoppers with an alcohol swab and let them dry. Draw 3 mL of bacteriostatic water and let it run slowly down the inside of the vial wall — not straight onto the powder.

  2. Hand gently swirling an upright vial in a small circle2

    Swirl gently

Swirl or roll the vial gently until the powder has dissolved. Give it a few minutes to settle if needed.

  3. A vial being shaken with a red cross through it3

    Don't shake it

Shaking can damage the peptide and cause foaming. Gentle swirling only.

  4. Close-up of a vial with small undissolved particles and a caution symbol4

    Check it's fully clear

Only use it once it is fully clear with no particles. Label the vial with the date and 1.67 mg/mL, then store it in the fridge at 2–8 °C.

How it works

MGF is a term used for an IGF-1 splice variant, often called IGF-1Ec in humans. A small human exercise experiment found MGF messenger RNA rose 2.5 hours after resistance exercise in younger participants, but not older participants.

A synthetic 24-amino-acid MGF E-peptide has been studied in cells and animals. A review notes that an equivalent endogenous peptide has not been identified in cultured cells, animal tissues or biological fluids, so natural IGF-1 gene expression does not establish that injected synthetic peptide acts the same way.

Laboratory findings are inconsistent: one primary human-cell experiment reported increased satellite-cell activation and fusion, whereas another experiment found no proliferative effect at concentrations up to 500 ng/mL.

Illustrated mechanism explainer

What the research shows

  • In a human expression experiment, 8 young men and 7 older men completed 10 sets of 6 knee extensions at 80% of one-repetition maximum. Quadriceps samples 2.5 hours later showed a significant MGF mRNA rise in the younger group but not the older group; this did not administer MGF peptide.
  • In a rabbit bone-defect experiment, 3 groups of 9 received 0, 28.5 or 57 mcg/kg MGF-Ct24E for 5 days. At 8 weeks, 4 of 9 high-dose defects had radiographic healing versus 0 of 9 in either control or low-dose groups; animal results do not establish a human dose.
  • Primary human muscle-cell cultures exposed to MGF-E reported increased proliferation and fusion in neonatal and young-adult cells, but another in-vitro experiment found no increase in human myoblast proliferation with MGF peptide concentrations up to 500 ng/mL.
  • The TGA ARTG search for ‘Mechano Growth Factor’ returned no matching results. MGF is not an approved medicine in Australia, and no FDA-approved MGF medicine label is cited here.

Reported side effects

  • Published evidence reviewed here does not provide a human adverse-event rate or long-term safety profile for administered MGF, so risks cannot be quantified.
  • FDA says the related PEG-MGF formulation has no human exposure data and may pose immunogenicity risks from aggregation, peptide-related impurities and active-ingredient characterisation. This is a warning about PEG-MGF, not a measured risk rate for non-PEG MGF.

Important notes

  • MGF is not an approved medicine in Australia: the ARTG search for ‘Mechano Growth Factor’ returned no matching results. No FDA-approved MGF medicine label is cited here.
  • Prohibited in sport: WADA identifies MGFs as explicitly mentioned on its Prohibited List, and its current S2 growth-factor category includes IGF-1 and its analogues.
  • The amounts above are copied from an educational reference protocol, not from a clinically established MGF dose. Synthetic MGF findings are inconsistent and do not demonstrate muscle repair or performance benefit in people.
  • Never use a mixed vial that is cloudy or has particles in it.

Injection basics

  • Use a new sterile 1 mL (100-unit) U-100 insulin syringe, 27–31G, every time — never reuse or share.
  • Wipe the vial stopper and the skin with an alcohol swab and let both dry first.
  • Inject under the skin (subcutaneously) of the abdomen, thigh or upper arm, and rotate sites.
  • Put used syringes straight into an approved sharps container — the NDSS lists Australian disposal options.

06 · QUICK ANSWERS

MGF questions

How many units is 100 mcg of MGF?

With 3 mL in a 5 mg vial, the concentration is 1.67 mg/mL. A 100 mcg amount is 6 units on a U-100 syringe, which is 0.06 mL.

How much bacteriostatic water for a 5 mg MGF vial?

The cited 5 mg reference protocol uses 3 mL. That calculation gives 1.67 mg/mL, or about 16.7 mcg per U-100 syringe unit.

How many units is 300 mcg of MGF?

With the default 5 mg plus 3 mL mix, 300 mcg is 18 units on a U-100 syringe, equal to 0.18 mL.

How long does a 5 mg MGF vial last?

Ignoring preparation losses, a 5 mg vial contains 50 daily 100 mcg amounts, 25 daily 200 mcg amounts or about 16.7 daily 300 mcg amounts. The 3 mL default mix changes the volume drawn, not the total peptide amount.
