Oxytocin Dosage Protocol

OXT · neuropeptide hormone and oxytocin-receptor ligand

The referenced 10 mg schedule uses 100 mcg once daily in weeks 1–2, increasing by 100 mcg every 2 weeks to 500 mcg in weeks 9–12. The default 3 mL mix makes 3.33 mg/mL, so the 100 mcg start is 3 units (0.03 mL) on a U-100 syringe.

The reference schedule increases by 100 mcg every 2 weeks through week 12.

The chart is for subcutaneous measurement only; intranasal, intravenous and obstetric oxytocin amounts are not interchangeable.

The reference page describes an 8–12 week daily schedule and mentions an optional extension to 16 weeks.

02 · HOW TO MIX

Mixing a 10 mg vial with 3 mL of BAC water

Using a 1 mL (100-unit) insulin syringe, 3 full draws of bacteriostatic water gives 3.33 mg/mL.

  1. Bacteriostatic water running slowly down the inside wall of a peptide vial1

    Add the water slowly

Wipe both rubber stoppers with an alcohol swab and let them dry. Draw 3 mL of bacteriostatic water and let it run slowly down the inside of the vial wall — not straight onto the powder.

  2. Hand gently swirling an upright vial in a small circle2

    Swirl gently

Swirl or roll the vial gently until the powder has dissolved. Give it a few minutes to settle if needed.

  3. A vial being shaken with a red cross through it3

    Don't shake it

Shaking can damage the peptide and cause foaming. Gentle swirling only.

  4. Close-up of a vial with small undissolved particles and a caution symbol4

    Check it's fully clear

Only use it once it is fully clear with no particles. Label the vial with the date and 3.33 mg/mL, then store it in the fridge at 2–8 °C.

How it works

Oxytocin is a naturally occurring nine-amino-acid peptide hormone. It binds the oxytocin receptor, a G-protein-coupled receptor that signals mainly through Gq/11 and phospholipase C pathways.

Its established physiological roles include uterine contraction and milk ejection. Research also examines oxytocin signalling in social behaviour, stress, pain and appetite, but findings depend on the formulation, route, population and outcome measured.

What the research shows

  • The TGA ARTG lists SYNTOCINON oxytocin 10 IU/1 mL injection ampoule as a registered medicine (ARTG 13383); this regulated finished injection is not the same product as a freeze-dried 10 mg research vial.
  • The US oxytocin injection label uses an initial intravenous infusion of 0.5–1 milliunit/minute for medically indicated labour induction, with 1–2 milliunit/minute increases at 30–60 minute intervals under hospital monitoring; these obstetric directions are not this chart.
  • In a randomised, double-blind social-anxiety experiment, 25 participants received 24 IU intranasal oxytocin or placebo with exposure therapy; positive self-evaluations improved, but overall symptom outcomes did not differ between groups.
  • A 2022 review describes oxytocin receptor signalling and research across several conditions, while emphasising that further investigation is needed before new therapies can be established.

Reported side effects

  • The medical-injection label reports maternal nausea, vomiting, hypertensive episodes, cardiac arrhythmias and anaphylactic reactions; its safety information is from obstetric care, not this research-vial schedule.
  • Excess oxytocin exposure in the obstetric setting can cause uterine hypertonicity, spasm or rupture, and can harm the fetus or newborn; labour use requires continuous medical supervision.
  • The label reports severe water intoxication with convulsions and coma after a slow 24-hour infusion, a route and setting that are not comparable with this chart.
  • Repeated subcutaneous use of this freeze-dried research vial does not have an established clinical safety profile; Peak describes its product as in-vitro research material, not for human use.

Important notes

  • Syntocinon is a TGA-registered oxytocin 10 IU/1 mL injection ampoule; Peak's 10 mg freeze-dried research vial is a different product and Peak states it is for in-vitro research only.
  • The FDA label says oxytocin for labour induction or augmentation must be given intravenously with adequate medical supervision in a hospital; do not use the research chart for obstetric care.
  • Intranasal oxytocin studies use device-delivered international units, while this reference chart is subcutaneous microgram measurement; the routes and formulations are not interchangeable.
  • The current 2026 WADA Prohibited List was checked and does not name oxytocin; athletes should still check their sport's current anti-doping advice.
  • Never use a mixed vial that is cloudy, discoloured or has particles in it.

Injection basics

  • Use a new sterile 1 mL (100-unit) U-100 insulin syringe, 27–31G, every time — never reuse or share.
  • Wipe the vial stopper and the skin with an alcohol swab and let both dry first.
  • Inject under the skin (subcutaneously) of the abdomen, thigh or upper arm, and rotate sites.
  • Put used syringes straight into an approved sharps container — the NDSS lists Australian disposal options.

06 · QUICK ANSWERS

Oxytocin questions

How many units is 100 mcg of oxytocin from a 10 mg vial?

With the default 3 mL mix, the concentration is 3.33 mg/mL. A 100 mcg amount is 3 U-100 units, or 0.03 mL.

How many units is 500 mcg of oxytocin?

With 3 mL in a 10 mg vial, 500 mcg is 15 U-100 units, or 0.15 mL. This is the week 9–12 amount in the attributed reference chart.

How much BAC water is used for an oxytocin 10 mg vial?

The default and attributed reference mix is 3 mL, which gives 3.33 mg/mL. At that concentration, 1 U-100 unit is 33.33 mcg and 30 units is 1 mg.

How long does a 10 mg oxytocin vial last?

The theoretical yield is 100 daily amounts at 100 mcg, 50 at 200 mcg, about 33 at 300 mcg, 25 at 400 mcg and 20 at 500 mcg. Handling loss and the product's storage period can shorten this.

Is a 10 mg research vial the same as Syntocinon?

No. The TGA lists Syntocinon as a registered 10 IU/1 mL oxytocin injection ampoule. Peak states its 10 mg freeze-dried vial is for in-vitro research only, so the products, routes and controls are different.
