NAD+ Dosage Protocol

Nicotinamide adenine dinucleotide · coenzyme, not a peptide

The reference calculation schedule uses 50 mg once daily in week 1, 75 mg in week 2, then 100 mg once daily through week 16. With Peak's 5 mL option, a 1000 mg vial is 200 mg/mL, so the 50 mg starting amount is 25 units (0.25 mL) on a U-100 syringe.

02 · HOW TO MIX

Mixing a 1000 mg vial with 5 mL of BAC water

Using a 1 mL (100-unit) insulin syringe, 5 full draws of bacteriostatic water gives 200 mg/mL.

  1. Bacteriostatic water running slowly down the inside wall of a peptide vial1

    Add the water slowly

Wipe both rubber stoppers with an alcohol swab and let them dry. Draw 5 mL of bacteriostatic water and let it run slowly down the inside of the vial wall — not straight onto the powder.

  2. Hand gently swirling an upright vial in a small circle2

    Swirl gently

Swirl or roll the vial gently until the powder has dissolved. Give it a few minutes to settle if needed.

  3. A vial being shaken with a red cross through it3

    Don't shake it

Shaking can damage the peptide and cause foaming. Gentle swirling only.

  4. Close-up of a vial with small undissolved particles and a caution symbol4

    Check it's fully clear

Only use it once it is fully clear with no particles. Label the vial with the date and 200 mg/mL, then store it in the fridge at 2–8 °C.

Keep the unopened powder dry, dark and cold; Peak states 2–8 °C, or −20 °C for long-term storage.
After mixing
Store at 2–8 °C away from light; Peak says to keep the vial upright and use within about 28 days.

Do not freeze the prepared solution or use it if it is cloudy, discoloured or has particles.

How it works

NAD+ is the oxidised form of nicotinamide adenine dinucleotide, a coenzyme rather than a peptide. It transfers electrons in cellular redox reactions and cycles with NADH in energy metabolism.

It is also used by enzymes including sirtuins and PARPs, linking NAD biology with signalling and DNA-repair research. These mechanisms do not establish a health benefit or a human injection dose.

Illustrated mechanism explainer

What the research shows

  • In a human pharmacokinetic pilot, 8 men received 750 mg NAD+ intravenously over 6 hours and 3 received saline; plasma NAD+ rose 398% at the end of infusion. This was supervised IV research, not a daily subcutaneous schedule.
  • The same 11-person pilot reported no observed adverse events during the 6-hour NAD+ or saline infusion; it was too small and used a different route to establish safety of the charted amounts.
  • A 2024 systematic review included 10 randomised studies with 489 participants across varied conditions. Its conclusions chiefly concerned oral NADH and precursors, not direct NAD+ injections.
  • A completed 36-person registered long-COVID project combined oral low-dose naltrexone with a 400 mg NAD+ iontophoresis patch once weekly for 12 weeks. It does not validate this chart's route, frequency or amounts.
  • The TGA says NAD and NAD+ are not permitted ingredients in listed medicines in Australia; goods outside the ARTG have not been evaluated by the TGA for quality, safety or effectiveness.

Reported side effects

  • Direct human evidence for repeated subcutaneous NAD+ is limited. The 11-person IV pilot found no observed adverse events during its 6-hour observation period, which cannot establish safety for another route or schedule.
  • Across the 10 studies in a systematic review of NAD, NADH and related interventions, reported effects included muscle pain, nervous-system symptoms, fatigue, sleep disturbance and headache; the studies used varied formulations and populations.
  • Injection-specific tolerability, longer-term risks and product quality for this community calculation schedule have not been established in controlled human research.

Important notes

  • Peak and Pure describe their linked 1000 mg products as research-only lyophilised powders and not for human consumption.
  • The TGA states NAD and NAD+ are not permitted ingredients in listed medicines in Australia. Do not treat the chart as evidence that a product is TGA approved, safe or effective.
  • The listed amounts are from a community-reported calculation page, not an approved label or a clinically established human subcutaneous dose.
  • The 2026 WADA Prohibited List does not name NAD+; athletes should obtain advice from their anti-doping organisation because WADA says the list and its categories must be checked for the specific substance and method.
  • Never use a mixed vial that is cloudy or has particles in it.

Injection basics

  • Use a new sterile 1 mL (100-unit) U-100 insulin syringe, 27–31G, every time — never reuse or share.
  • Wipe the vial stopper and the skin with an alcohol swab and let both dry first.
  • Inject under the skin (subcutaneously) of the abdomen, thigh or upper arm, and rotate sites.
  • Put used syringes straight into an approved sharps container — the NDSS lists Australian disposal options.

06 · QUICK ANSWERS

NAD+ questions

How many units is 50 mg of NAD+ in a 1000 mg vial?

With the default 5 mL mix, the concentration is 200 mg/mL. A 50 mg amount is 0.25 mL, or 25 units on a U-100 syringe.

How many units are 75 mg and 100 mg of NAD+?

At the default 200 mg/mL concentration, 75 mg is 0.375 mL or 37.5 U-100 units. One hundred milligrams is 0.5 mL or 50 U-100 units.

How much BAC water is used for NAD+ 1000 mg?

This guide defaults to Peak's highlighted 5 mL option, which produces 200 mg/mL. Peak also lists 3 mL and 10 mL calculation options; the reference protocol uses 3 mL, not the 5 mL default used here.

How long does a 1000 mg NAD+ vial last?

At a constant 50 mg daily it contains 20 days of material, at 75 mg daily about 13.3 days, and at 100 mg daily 10 days. The full reference calculation schedule totals 5,075 mg over 8 weeks, 7,875 mg over 12 weeks and 10,675 mg over 16 weeks.

Is there an established daily NAD+ injection dose?

No. The chart source calls its 50–100 mg daily amounts a community-reported calculation pattern. Published direct NAD+ work is limited and includes a supervised 750 mg intravenous infusion, which does not establish a subcutaneous dose.
