DSIP Dosage Protocol

Delta sleep-inducing peptide · emideltide nonapeptide

The reference protocol lists 100 mcg nightly in week 1, then 150 mcg in week 2, 200 mcg in week 3 and 250–300 mcg for weeks 4–8. With 3 mL of bacteriostatic water, a 10 mg vial is 3.33 mg/mL, so the 100 mcg starting amount is 3 units on a U-100 syringe.

02 · HOW TO MIX

Mixing a 10 mg vial with 3 mL of BAC water

Using a 1 mL (100-unit) insulin syringe, 3 full draws of bacteriostatic water gives 3.33 mg/mL.

  1. Bacteriostatic water running slowly down the inside wall of a peptide vial1

    Add the water slowly

Wipe both rubber stoppers with an alcohol swab and let them dry. Draw 3 mL of bacteriostatic water and let it run slowly down the inside of the vial wall — not straight onto the powder.

  2. Hand gently swirling an upright vial in a small circle2

    Swirl gently

Swirl or roll the vial gently until the powder has dissolved. Give it a few minutes to settle if needed.

  3. A vial being shaken with a red cross through it3

    Don't shake it

Shaking can damage the peptide and cause foaming. Gentle swirling only.

  4. Close-up of a vial with small undissolved particles and a caution symbol4

    Check it's fully clear

Only use it once it is fully clear with no particles. Label the vial with the date and 3.33 mg/mL, then store it in the fridge at 2–8 °C.

How it works

DSIP, also called emideltide, is a nine-amino-acid peptide historically investigated in sleep physiology and neuroendocrine research. Its name reflects early animal work rather than an established treatment effect.

The proposed biology remains unresolved. A 2006 review found that the DSIP gene, precursor protein and possible related receptor had not been isolated, and described the sleep-factor hypothesis as weakly documented.

Illustrated mechanism explainer

What the research shows

  • In a 1981 double-blind crossover experiment, six healthy volunteers received slow intravenous DSIP at 25 nmol/kg; median total sleep time during a 130-minute observation interval was 59% higher than with placebo. This was intravenous research, not the subcutaneous schedule shown above.
  • A 1992 double-blind study in 16 people with chronic insomnia used intravenous DSIP at 25 nmol/kg before three nights. Objective sleep efficiency was higher and sleep latency shorter than placebo, but the authors judged the effects weak and found no change in subjective sleep quality.
  • FDA's 2026 assessment found no data supporting emideltide effectiveness for chronic insomnia, narcolepsy or opioid withdrawal by the proposed subcutaneous route, and no clinical safety data for that route.
  • Neither emideltide free base nor emideltide acetate is a component of an FDA-approved drug. Searches of the public ARTG for DSIP, emideltide and delta sleep-inducing peptide returned no matching record when checked on 25 October 2026.
  • A ClinicalTrials.gov intervention search for delta sleep-inducing peptide, DSIP or emideltide returned no records when checked on 25 October 2026.

Reported side effects

  • There are no clinical safety data for the proposed subcutaneous route in FDA's emideltide assessment, so its risks for that route are not established.
  • FDA reported transient headache, nausea and vertigo as common side effects in older intravenous withdrawal reports, with cases of progressive hypotension after the second intravenous injection; withdrawal symptoms make interpretation difficult.
  • FDA also identifies potential injectable-peptide risks from aggregation, peptide-related impurities and immunogenicity.

Important notes

  • DSIP is not an FDA-approved medicine, and no matching DSIP or emideltide record appeared in the public ARTG search on 25 October 2026. Both listed suppliers describe their 10 mg product as research material, not for human consumption.
  • Prohibited in sport: DSIP falls within WADA S0 for pharmacological substances without current approval by a governmental regulatory health authority for human therapeutic use; WADA lists S0 as prohibited at all times.
  • FDA concluded that subcutaneous emideltide safety and effectiveness data are insufficient and highlighted possible immunogenicity and impurity-related risks.
  • Never use a mixed vial that is cloudy or has particles in it.

Injection basics

  • Use a new sterile 1 mL (100-unit) U-100 insulin syringe, 27–31G, every time — never reuse or share.
  • Wipe the vial stopper and the skin with an alcohol swab and let both dry first.
  • Inject under the skin (subcutaneously) of the abdomen, thigh or upper arm, and rotate sites.
  • Put used syringes straight into an approved sharps container — the NDSS lists Australian disposal options.

06 · QUICK ANSWERS

DSIP questions

How many units is 100 mcg of DSIP?

With the default 3 mL mix in a nominal 10 mg vial, the concentration is 3.33 mg/mL. A 100 mcg amount is 3 units on a U-100 syringe, or 0.03 mL.

How many units is 250 mcg of DSIP?

With the default 10 mg plus 3 mL mix, 250 mcg is 7.5 units on a U-100 syringe, which is 0.075 mL.

How much bacteriostatic water is used for DSIP 10 mg?

The default mix is 3 mL because Peak's DSIP page and the attributed reference protocol use that volume. It produces 3.33 mg/mL, where 30 U-100 units equals 1 mg or 0.30 mL.

How long does a 10 mg DSIP vial last?

A nominal 10 mg vial contains 100 amounts of 100 mcg, 50 amounts of 200 mcg, or about 33 amounts of 300 mcg. The number of days depends on the attributed step-up schedule and any material lost during preparation.
