Triple Agonist (GLP-1/GIP/Glucagon)
Also available: 20 mg vial
IBI362 / LY3305677 · dual GLP-1 and glucagon receptor agonist
Published GLORY-1 escalation was once weekly: 2 mg in weeks 1–4, 4 mg in weeks 5–8, then 6 mg. With 1 mL in a 10 mg vial, the mix is 10 mg/mL, so the 2 mg starting amount is 20 units (0.2 mL) on a U-100 syringe.

Published weekly escalation · 10 mg vial + 1 mL BAC water
1 mL is the default calculator mix because it makes the 2 mg, 4 mg and 6 mg chart amounts 20, 40 and 60 units.
| Week | Weekly dose | Syringe units | Syringe fill |
|---|---|---|---|
| Weeks 1–4Start | 2 mg | 20 units0.2 mL | |
| Weeks 5–8 | 4 mg | 40 units0.4 mL | |
| Week 9 onwardsMaintenance | 6 mg | 60 units0.6 mL |
Once weekly · subcutaneous injection
The published schedule steps up after 4 weeks at each lower amount.
The chart stops at 6 mg, the highest amount in this published escalation.
This chart reproduces a published clinical-trial escalation; mazdutide has no TGA or US FDA approval. Education only, not medical advice. Dose amounts adapted from GLORY-1 published 6 mg dose-escalation schedule (Ji et al., New England Journal of Medicine, 2025); units are recalculated for the vial and mix selected above.
Using a 1 mL (100-unit) insulin syringe, one full draw of bacteriostatic water gives 10 mg/mL.
1Wipe both rubber stoppers with an alcohol swab and let them dry. Draw 1 mL of bacteriostatic water and let it run slowly down the inside of the vial wall — not straight onto the powder.
2Swirl or roll the vial gently until the powder has dissolved. Give it a few minutes to settle if needed.
3Shaking can damage the peptide and cause foaming. Gentle swirling only.
4Only use it once it is fully clear with no particles. Label the vial with the date and 10 mg/mL, then store it in the fridge at 2–8 °C.
Counts follow the dosage chart above at the highest amount in each step, using a 10 mg vial mixed with 1 mL.
Plus an approved sharps container. Syringes and swabs are sold by Australian pharmacies — Peak Peptides and Pure Performance supply the bacteriostatic water.

Mazdutide, also called IBI362 or LY3305677, is an oxyntomodulin analogue that activates both the GLP-1 receptor and the glucagon receptor. It is given once weekly in the published obesity research program.
GLP-1 receptor activity is associated with glucose-dependent insulin release, slower gastric emptying and appetite-related effects. Glucagon receptor activity may affect hepatic energy metabolism and energy expenditure; the combined mechanism is being investigated in clinical research.

With 1 mL in a 10 mg vial, the concentration is 10 mg/mL. The 2 mg, 4 mg and 6 mg amounts are 20 units (0.2 mL), 40 units (0.4 mL) and 60 units (0.6 mL) on a U-100 syringe.
This calculator's default is 1 mL for a 10 mg/mL concentration. At that concentration, 10 units is 1 mg and 20 units is 2 mg. The selector also shows 1.5 mL and 2 mL options.
By mass alone, a 10 mg vial contains five 2 mg weekly amounts. At 4 mg it contains two full amounts with 2 mg remaining; at 6 mg it contains one full amount with 4 mg remaining. This arithmetic does not establish a validated mixed-vial expiry.
With 10 mg in 1 mL, 1 mg is 0.1 mL, which is 10 units on a U-100 syringe.
